Hello!
I want to set up time delays in the car.
I have an external USB audio interface and a measuring microphone.
I'm going to measure the arrival time of the signal from each speaker.
Should I use a loopback (connecting the output and input of the sound card)?
If it must be done, is there a connection diagram somewhere (figure)?

A timing reference is required for what you’ve got in mind. Acoustic timing also works in a car. I’ve used it. That said, which method is easiest in actual usage I think will depend on your car system, specifically how the signal will get to each channel and how easy it would be to maintain the same reference speaker in order to use acoustic timing.

For loopback, yes, interface channel 2 output to channel 2 input. Mic on channel 1 input. Channel 1 output to car audio input (however it can be done on your system). Disable all active processing on the interface when measuring.

Here’s a short video that shows how to set up an external interface with REW if you need it.

I may not understand your question…
Does your audio interface have 2 or more inputs and outputs?
The reference (loopback) channel is channel 2. The output is connected to the input.

One of the tweeters can be both measurable and reference.

It uses a loop to tie time. Instead of an additional speaker. Loop from the output of the sound card to one of the inputs of the sound card. This is possible, but it will be more accurate with an acoustic reference to time. That is, either an additional speaker(additional tweeter) or one of the tweeters to be measured.
